Oh come on. You might have a fair point if his reaction was something more generic like, "And I thought I knew everything about you!" But the very specific reference to sharing a hotel room is totally implying that Sherman Helmsley's character is grossed out by the idea of sleeping in such close proximity to this newly-revealed sissy.

 

Most men would be legitimately uncomfortable sleeping next to a transvestite. Especially men obsessed with being in in good public standing like Helmsley.

 

Bottom line: I bought that Sherman would be legitimately offended.
